Biochemical Effects of Mithramycin on Cultured Cells1

Following treatment with mithramycin, RNA synthesis was decreased by 85% in mouse embryo and BHK-21 cell cultures and by 45% in human conjunctiva cell cultures when compared to controls. The inhibition of RNA synthesis in BHK-21 cells was dosedependent and encompassed all cellular types of RNA. Although mithramycin inhibited protein and DNA syntheses in BHK-21 cells, these were not dose-related, which suggested that the primary site of action of this antibiotic was on RNA synthesis.
